I am having trouble updating my spysweeper. It say it cannot create the
file C:\Program FIles\Webroot\Spy Sweeper\Masters\masters.mst Access is
denied.
I have tried changing rights to the folder and file and shutting down the
service etc.. I still get this message. I also try right clicking and
running as administrator.

Spy Sweeper Version 5.3 is compatible with Windows Vista (according to their documentation).
